磷脂酶Cε1在结肠癌组织中的表达及意义

Expression and Significance of PI-PLCε1 in Colon Cancer

  • 摘要: 目的 通过检测结肠癌组织及其癌旁组织中磷酸肌醇特异性的磷脂酶Cε1 (PI-PLCε1)的表达情况，探讨PI-PLCε1在结肠癌发病过程中的临床意义。方法 采用实时荧光定量PCR（real-time quantitative PCR，qRT-PCR）及免疫组化的方法检测42例结肠癌组织及其相对应的癌旁组织中PI-PLCε1的表达情况，并分析肿瘤分化程度及肿瘤部位对于结肠癌组织中PI-PLCε1表达情况的影响。结果 qRT-PCR结果显示，结肠癌组织中PI-PLCε1基因的表达低于其癌旁组织（ P<0.05），不同肿瘤分化程度及肿瘤部位结肠癌组织中PI-PLCε1基因的表达差异没有统计学意义（ P>0.05）。免疫组化结果显示，结肠癌组织中PI-PLCε1蛋白的阳性表达率低于其癌旁组织（ P<0.05）。不同肿瘤分化程度结肠癌组织中PI-PLCε1蛋白的阳性表达率差异没有统计学意义（ P>0.05），但不同肿瘤部位中PI-PLCε1蛋白的阳性表达率差异有统计学意义（ P<0.05)。结论 PI-PLCε1在结肠癌组织中的表达显著减少，且与肿瘤分化程度无关。  
    Abstract: ObjectiveTo study the expression and clinical significance of phosphoinositide-specific phospholipase Cε1 (PI-PLCε1) in the pathogenesis of colon cancer. MethodsqRT-PCR and immunohistochemistry were used to detect the expression of PI-PLCε1in the 42 cases of colon cancer tissues and their corresponding adjacent tissues. And the effects of tumor differentiation and tumor site on the expression PI-PLCε1 of colon cancer tissues were compared. ResultsThe results of qRT-PCR showed that the expression of PI-PLCε1in colon cancer tissue significantly lower than that in the adjacent tissue ( P<0.05). The expression of PI-PLCε1gene of colon cancer tissue was not effected by tumor differentiation and tumor site ( P>0.05). The results of immuno-histochemistry showed that the positive expression rate of PI-PLCε1 protein in colon cancer tissue was significantly lower than that in the adjacent tissue ( P<0.05). The positive expression rate of PI-PLCε1 protein was not effected by tumor differentiation ( P>0.05), but the expression was different in tumor site ( P<0.05). Conclusion Expression of PI-PLCε1 was reduced in colon tissue and barely to tumor differentiation.
