芍药苷对MRL/lpr狼疮小鼠肺损伤的保护作用

The Study of Protective Effect of Paeoniflorin on Lung Injury in MRL/lpr Mice

  • 摘要: 目的 探讨芍药苷对MRL/lpr狼疮小鼠肺损伤的影响。 方法 将系统性红斑狼疮MRL/lpr小鼠随机分为MRL/lpr组、MRL/lpr+地塞米松 (1.5 mg/kg) 组,MRL/lpr+芍药苷(20 mg/kg)组,MRL/lpr+芍药苷 (40 mg/kg) 组, 每组10只,给药7 d;另取10只野生型C57BL/6小鼠作为正常对照组。第8天采用酶联免疫吸附法(ELISA)检测小鼠血清中超氧化物歧化酶(SOD)、丙二醛(MDA)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)和血清炎症因子白细胞介素6(IL-6)、白细胞介素1β(IL-1β)及肿瘤坏死因子α(TNF-α)水平;采用HE染色检测肺部病理改变,采用蛋白质免疫印迹(Western blot)技术检测各组小鼠肺组织中磷酸化磷脂酰肌醇-3激酶(P-PI3K)、磷酸化丝氨酸苏氨酸蛋白激酶(P-Akt)、磷酸化核因子κB p65(P-NF-κBp65)、磷酸化NF-κB抑制蛋白α(P-IκBα)蛋白的表达。 结果 芍药苷能显著降低血清MDA含量;提高血清SOD、CAT、GSH-PX水平,减少血清炎症因子水平,改善肺组织病理改变,并能显著抑制P-PI3K、P-Akt、P-NF-κBp65、P-IκBα蛋白表达。结论 芍药苷对MRL/lpr狼疮小鼠肺损伤起到保护作用。  
    Abstract: ObjectiveTo investigate the protective effects of paeoniflorin on the lung injury in systemic lupus erythematosus with mouse model. Methods Ten wild type mice and 40 MRL/lpr mice were used in this stuy. MRL/lpr mice were randomly assigned to MRL/lpr group, MRL/lpr + dexamethasone (1.5 mg/kg) group, MRL/lpr + paeoniflorin (20 mg/kg) group, and MRL/lpr + paeoniflorin (40 mg/kg). The levels of malondialdehyde(MDA),superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) in serum were detected. The serum levesl of inflammatory cytokines were measured. Lung pathological changes were determined by HE staining. The protein level of phospho-phosphatidylinositol-3 kinase (P-PI3K), phospho-serine-threonine kinase B(P-Akt), phospho-nuclear factor kappa B (P-NF-κB), phospho-inhibitor of nuclear factor kappa Bα (P-IκBα) were detected by Western blot. Results Paeoniflorin decreased serum level of MDA and increased the levels of SOD, CAT, GSH-PX, and decreased the levels of inflammatory cytokines. Paeoniflorin improved lung pathological changes and inhibited the protein levels of P-PI3K, P-Akt, P-NF-κBp65, and P-IκBα in the lung tissue of MRL/lpr mice. Conclusion Paeoniflorin may be beneficial for the prevention of lung injury in systemic lupus erythematosus.
